No it is not necessary.
Car air conditioning systems are like refrigerators, it is a sealed system.
The only reason it is necessary to replace the refrigerant is if there's been a leak and refrigerant is lost from the system or if there's been a mechanical failure in the system e.g. compressor, piping etc and then any remaining refrigerant in the system would have to be removed, the system repaired and then recharged with refrigerant and tested etc.
There's such a thing called deodourising the system so as to get rid of musty smells created by mould caught in the evaporator but this involves spraying the air intakes with an A/C disinfectant and running the system for a while. Also replacing the cabin air filter helps as well.
